 physical properties of amino acids plays a vital role in the structure of protein as proteins structure is made of arrangement of amino acids in such a manner that it can make the protein structure functional as per the desired function. 

explanation : 

Arrangement of amino acid is a physical property and if the arrangement is not as per the desired structure, the protein will not be functional as per the task. The amino acids may have an extra bond or additional bond in it self which changes its properties .Each protein has a unique structure that is folded in a unique form which makes the structure functional. The change in the amino acids structures may lead to misfunctioning of proteins.
